Identification of Porphyromonas gingivalis from the pocket with advanced periodontitis by PCR method

Abstract :

Objective  The purpose of this study was to confirm identification of Porphyromonas gingivalis by using the PCR method.

Materials and methods  Black pigmented Bacteroides (No.1, No.2 and No.3) were isolated from the pockets of three advanced periodontitis patients who came to Periodontal Department of Faculty of Dentistry, Chulalongkorn University for periodontal disease treatment. These bacterias were cultured on brain heart infusion-yeast extract blood (sheep) agar [BHI blood agar] in anaerobic chamber which contained 80% N2, 10% H2 and 10% CO2 at 37°C for 10 days. After these bacterias had already grown. DNA was isolated and amplified for using the PCR method.

Results  The result of this study showed that three types of bacteroides were isolated. Samples No.1 and No.2 had formed bands which were related to Primer Pg (19-+20) and Primer Pg (21+22). 

Conclusion  PCR method is the appropriate method to detect Pg from Periodontitis patient because it can detect Pg accurately and rapidly.
